Microscope Resolution

I have been interested in exploring various methods for measuring microscope resolution. I found a method using the Point Spread Function at 1/2 the maximum. The method uses sub micron fluorescent beads. https://imagejdocu.list.lu/_media/plugin/analysis/metroloj/metroloj.pdf It’s a plugin for ImageJ. Can this method be used for visible images. Could I use say 2um beads or smaller, image the beads then use this macro to get the PSF width at 1/2 maximum?

My goal would be to compare this method against the sharp edge method and the 1951 USAF target methods.

I would like to use these resolution methods to try to improve the resolution of the scope and possibly use them to evaluate a Super Resolution method like SIM on the openflexure system.
