I can’t seem to make focus stacking work with delta stage using imjoy.
Here’s an example of onion skin using all algorithms provided in imagej.
I suspect it is due to the Delta stage not being cartesian, hence moving z also moves the sample in the xy plane.
The colour correction also seems to be very off in those images.
There is clearly an xy shift between the different images. On the Delta stage it does need to move all motors for z motion, if you have moved sideways just before the z motion then the different backlash on the A, B,C motors will cause additional problems.
I would expect that imagej has options to align images before stacking, it would be a common need.
